Microsatellite DNA Somaclonal Variation of Regenerated Plants via Cotyledon Culture of Hot Pepper (Capsicum annuum L.)

In this study, microsatellite DNA markers were utilized to characterize somaclonal variation in regenerated plants from cultured cotyledons of Capsicum annuum L. cv Subicho. Twenty five microsatellite primer pairs were utilized in the screening of 41 regenerated plants derived from the cotyledons. Microsatellite DNA variation was detected among the regenerated plants at two SSR loci. The primer pairs for the SSR loci HpmsF15 and HpmsF037 revealed somaclonal variation in 5 out of the 41 plantlets (12%). However, 23 microsatellite markers evidenced no nuclear DNA polymorphisms. Nevertheless, the microsatellite marker technique can be successfully employed to assess genetic variations in somaclonal variants of the hot pepper.
